PX’Monoclonals offers services ranging from the antigen generation
through to antibody purification for research (immunohistochemistry,
FACS, ELISA assay, Western/Dot-blotting) or therapeutic purposes
(development of neutralizing antibodies).
The vocation of PX’Monoclonals is specifically to address antibodies development related issues such as immunization of animals with difficult to express antigens (use of genic immunization) or generation of mAbs directed against rare and weakly antigenic epitopes (use of substractive immunization).
According to the project specifications and objectives, the platform will propose the following steps:
• Production of complex immunogen if required
• Animal immunization
- from DNA, transfected
cells or purified protein
- use of standard or custom
(substractive) immunization protocols.
• Fusion et generation of hybridomas
• Screening of hybridomas
• Cloning of the hybridomas of interest by limited dilution
• Characterization of antibody
• Production and purification of mg scale antibody batch
Additional services
• Development of ELISA assay (ELISA-Sandwich method)
PX’Monoclonals proposes to develop ELISA assay for diagnostics applications and
in process or immuno-monitoring controls. Used as a research tool, ELISA assays
allow to assess the specificity and the affinity of the antibodies developed.
• Development of seroneutralizing test
The platform also proposes to develop cell based assays which aim at identifying
the neutralizing potential of the antibodies developed. For specific projects
involving dangerous or lethal pathogens, PX’Monoclonals will negotiate access
to P3/P4 laboratories located in Lyon.
